Etoposide prevents DNA topoisomerase II, thereby blocking DNA re-ligation. This causes detracting errors in DNA synthesis at the premycotic level of cell division and causes apoptosis of the tumor cell. The drug is cell cycle dependent and phase specific, affecting primarily the S and G2 phases of cell division. prohibition of the topoisomerase II alpha isoform occurs in the anti-tumour activity of etoposide. The drug is also capable of preventing the beta isoform, but inhibition of this target is not analogue with the anticancer activity. It is instead along with the carcinogenic effect.
